Corneal Epithelial Stem Cell and Disease – Past, Present and Future

چکیده

Differential microarray experiments have shown that there are 186 transcripts predominantly overexpressed in the limbus, 644 in the cornea and 506 in the conjunctiva of the vervet monkey. In humans, 332 and 592 transcripts are predominantly overexpressed in the conjunctival and corneal epithelia, respectively. Mounting evidence generated from clinical observations and experimental animal studies over the last three decades strongly suggests that corneal epithelial stem cells locate at the limbus. More specifically, the interpalisadal epithelial rete ridges in the palisades of Vogt may serve as a repository for limbal stem cells (LSCs).

Comparison of Ultra Structure and Gene Expression of Cultured Limbal Stem Cells and Fresh Conjunctival, Limbal and Corneal Tissues

Purpose: The present study intends to show the characteristics of cultured limbal stem cell (CLSCs) and to compare them with normal Conjunctival (C), Limbal (L) and Cornea (K) tissues. Materials and Methods: The expressions of a set of genes potentially involved in differentiation and stemness function of limbal stem cells were assessed in freshly prepared limbal, corneal, and conjunctival tis...

Variations of the Normal Human Limbal Stem Cells Detected by In Vivo Confocal Microscopy

Background  To report normal variations of the limbal structures using in vivo laser scanning confocal microscopy. Methods: This was a retrospective study of fourteen eyes from 11 healthy individuals. Confocal imaging of cornea and limbus was performed using Heidelberg Retina Tomograph III Rostock Corneal Module. Results: The typical structure of the palisades of Vogt (POV) was detected ...

Corneal epithelial stem cells: past, present, and future.

Corneal epithelium is a self-renewing tissue. Recent studies indicate that corneal epithelial stem cells reside preferentially in the basal layer of peripheral cornea in the limbal zone, rather than uniformly in the entire corneal epithelium. This idea is supported by a unique limbal/corneal expression pattern of the K3 keratin marker for corneal-type differentiation; the preferential distribut...

Hematopoietic stem cell transplant therapy, clinical trials, complications, and quality of life for patients with Sickle cell anemia: Clinical potential and future perspectives

Background: Sickle cell anemia (SCA) is an inherited monogenic disorder. The clinical symptoms of SCA are protean, including vaso-occlusion, hemolysis, early stroke, leg ulcers, multi-organ failure, and increased risk of premature death. Hematopoietic stem cell transplantation is the only treatment identified to reduce SCA-related organ damage. Unfortunately, graft rejection is a significant im...

Corneal epithelial stem cells at the limbus: looking at some old problems from a new angle.

Corneal epithelium is traditionally thought to be a self-sufficient, self-renewing tissue implying that its stem cells are located in its basal cell layer. Recent studies indicate however that corneal epithelial stem cells reside in the basal layer of peripheral cornea in the limbal zone, and that corneal and conjunctival epithelia represent distinct cell lineages. These ideas are supported by ...
